Semaglutide: A Research Guide
Semaglutide is the most extensively studied weight-loss compound in clinical literature, with outcomes documented across more than 17,000 trial participants in the STEP and SELECT trial programs. It is also the first compound in its class shown to reduce major cardiovascular events independent of weight loss.

Mechanism
As a GLP-1 receptor agonist, semaglutide regulates appetite and glucose metabolism centrally and peripherally. Neuroimaging studies found reduced activation of brain reward centers in response to high-calorie food cues, suggesting a genuine shift in food preference rather than simple appetite suppression. That distinction may explain the durability of weight loss observed at the two-year mark in extension studies.
